Abstract

Pluchea indica L. is a plant containing sterols, flavonoids, tannins, and hydroquinone phenols. Flavonoid and phenolic can be extracted with percolation, maceration, soxhlet, and reflux methods. This study aims were to compare the total flavonoid and phenolic content of methanolic extract of Pluchea indica L. leaves (MEPL) in various methods of extraction. Determination of total flavonoids and phenolic content was done by spectrophotometry. The reagents of AlCl 3 and Folin-ciocalteu were used in these spectrophotometry method with quercetin and gallic acid as a standard. The serial concentrations of quercetin were 5, 10, 15, 20 and 25 μg/mL at λ = 428.90 nm with operating time 30 min. The equation of standard curve was y = 0.02001x + 0.10906 (x = total flavonoid content in μg/mL, y = absorbance) with r = 0.99848. Serial concentrations of gallic acid were 10, 20, 30, 40 and 50 μg/mL at λ = 757.80 nm with operating time 2 hours. The equation of standard curve was y = 0.01086 x + 0.12030 (x = total phenolic content in μg/mL, y = absorbance) with r = 0.99808. Total flavonoid and phenolic content in MEPL from various extraction methods were analyzed with One Way Anova statistic followed by Bonferroni test at 95% confidence level. Result showed that the total flavonoid content in MEPL by percolation, maceration, soxhlet and reflux method were 66.75; 51.80; 47.72 and 38.39 mg/gram extract. The total phenolic was respectively 116.95; 84.11; 67.53 and 44.36 mg/gram extract. Statistical results showed significant differences in the total flavonoids and phenolic content among the various extraction methods.
