Abstract
Water and ethanol extract of long bean (Vigna unguiculata (L) Walp) seed have antibacterial activity against Bacillus subtilis and Escherichia coli. Water extract contain flavonoid, glycoside, alkaloid, tannin, saponin and polyphenol, while ethanol extract contain carbohydrate, alkaloid, glycoside, saponin, steroid, tannin and polyphenol. This study aims were to compare antibacterial activity of water fraction derived from water and ethanol extract long bean seed to Bacillus subtilis and Escherichia coli. This research is an experimental research with post-test only control group design. Long bean seed were extracted using two methods, wheter infundation using water solvent and soxhletation using 96% ethanol, and then the solvent evaporated using rotary evaporator. The water and ethanol extract were partitioned using n-hexane and water. Water fractions were tested for their antibacterial activity by dilution method at concentration series (2000, 2250, 2500, 2750, and 3000 μg/ml) with positive control of tetracycline HCl. The lowest concentration that can inhibit the growth of test bacteria is called MIC (Minimum Inhibitory Concentration) characterized by clearly appearance in the test tube. The results showed difference MIC value between Bacillus subtilis and Escherichia coli. MIC value of water fraction from water and ethanol extract long bean seed is 2250 μg/ml againts B. subtilis. Meanwhile, MIC value of water fraction from water extract is 2500 μg/ml, whereas from water fraction of ethanol extract is 2750 μg/ml againts E. coli.
