Abstract

Sapodilla (Manilkara zapota L.) leaves has a chemical constituents, one of them was flavonoid, have bioactivity as antioxidants, antibacterial, antitumor, antiartritis and antiemetik. Flavonoid could be extracted by maceration, percolation, reflux and soxhletation method. The purpose of this study was to compared the content of total flavonoid from the ethanol extract of sapodilla leaves among the extraction methods. The proccess of extraction using ethanol as a solvent. Determination of total flavonoid was performed by spectrophotometric method using alumunium chloride (AlCl 3 ) and quercetin as a standard. Quercetin measured by spectrophotometric UV-Vis at serial concentrations of 2, 4, 6, 8, 10, and 12 μg/mL at λ 436,40 nm with an operating time for 30 minutes. Standard curve equation of quercetin were y = 0,04069x+0,11019 (x = total flavonoids content in μg/gram, y = absorbance) with r value = 0,99820. Total flavonoids content of ethanol extract sapodilla leaves among the extraction methods was analyzed using statistical One Way Anova then continued by Bonferonnitest with 95% confidence level. The results showed that the content of total flavonoids in the ethanol extract of sapodilla leaves with maceration method, percolation, soxhletation, and reflux were 43.79; 64.88; 49.41; and 32.18 mg/gram respectively. The statistical results showed the significant differences of total flavonoid content. The exctraction method that produces the highest levels of total flavonoids content is percolation.
