Abstract

Cisplatin chemotherapy agents in the treatment of cancer have some side effects and resistance, so it needs to be done to increase the effectiveness and reduce the side effects, it was with combination therapy using natural ingredients. Panduratin A was one of the compounds contained in the temu kunci rhizhome that has been shown to have cytotoxic activity against some cancer cells. The purpose of this research was to know the synergism cytotoxic effect of ethanolic extract temu kunci rhizhome (EERTK) and cisplatin on cancer cell of serviks HeLa. This type of research was laboratory experimental. The EERTK was extracted by maceration method using 70% ethanol solvent, and thickened using rotary evaporator. The cytotoxic test of EERTK for a single or a combination concoction with cisplatin was done by MTT assay method. The absorbance data that has been obtained was converted into the percentage of living cells.The data analysis using SPSS 16 for Windows pro analysis between the percentage of living cells and concentration to obtain IC50 values. The combined index (CI) was calculated to determine whether the resulting effect was synergistic, additive or antagonistic. The results of a combination of ethanolic extracts of temu kunci rhizome with cisplatin resulted in a synergistic effect on the ratio of EERTK IC50 (18.78 μg/ml) – cisplatin IC50 (16.745 μM) and combination EERTK IC50 (37.56 μg/ml) and all concentrations of cisplatin with CI values < 0.9.
