Abstract

Cortex of rambutan is a part of the plant are used by people to treat various diseases. Flavonoids and phenolic in rambutan proven cortex can be used as an antifungal and antioxidants. This study aims to know the methods of assay validation test flavonoids and total phenolic in ethyl acetate fraction of ethanol extract from the cortex of rambutan (Nephelium lappeceum L.) Extraction is done by percolation method using ethanol 96%, followed by fractionation method using a solvent ethyl acetate. Identification of the active compound and phenolic flavonoid phytochemical test done. Levels of flavonoids and phenolic total measured using colorimetric methods using magnesium powder reagent Mg 0.1 and 0.4 mL of amyl alcohol (flavonoids) and Folin ciocalteau (phenolic) and then measure the absorbance is read using UV -Vis. Test validation is performed by various tests, among others: precision test, test , test accuracy, linearity test, test LOD and LOQ followed by assay of flavonoids and total phenolic in ethyl acetate fraction of ethanol extract from the cortex of rambutan and subsequent data analysis using linear regression. The results of this study indicate that ethyl acetate fraction ethanol extract of rambutan stem cortex contains flavonoids and phenolic. The average yield assay of phenolic compounds and flavonoids are 23.826%, 0.151%
