Abstract

Failure treatment of cancer therapy encourages the development of anticancer drugs from natural materials. Kluwak contains tocopherol and tokotrienol, that are suspected of having anticancer activity. This study aims to develop the potential of natural ingredients from kluwak seeds as cytotoxic agents against T47D breast cancer cells and prove that ultrasonic method has better cytotoxic activity compared with maceration method. Extraction of kluwak seed powder by maceration and ultrasonic method using 70% ethanol solution. The filtrate obtained was concentrated with rotary evaporator. Followed by fractionation process using n-hexane solvent. The cytotoxicity test was performed with an MTT assay using five grade levels of 250; 300; 400; 500 and 750 μg / ml with an IC 50 parameter showing the concentration of the test solution causing the death of 50% of the cell population. IC 50 was analyzed using linear regression. Comparison of extraction methods to cytotoxic activity was analyzed using SPSS with One Way ANOVA. The n-hexane fraction of ethanol extract of kluwak seed by maceration and ultrasounic method was shown to have cytotoxic activity against T47D breast cancer cells with IC 50 value of 664 μg / ml and 461,75 μg / ml. The nhexane fraction of ethanol extract of kluwak seed with ultrasonic method has greater cytotoxic activity than the maceration method.
